Object of research: difficultly propagated clematis varieties .

Aim of work: to create a collection of aseptic cultures of hard-to-breed clematis varieties; to obtain aseptic cultures of hard-to-multiply clematis varieties; to select the optimal hormonal composition of the nutrient medium MurasigeSkuga for mass multiplication of clematis in vitro and for accelerated proliferation of axillary meristems .

Research methods: the method of microclonal propagation of plants, microcirculation .

In the course of the work, a collection of aseptic cultures of hard-to-breed clematis species was obtained: 'Danuta', 'Mazury', 'Fujimusume', 'Marmori', 'Kakio', 'William Kennett', 'Carmencita', 'Westerplatte', 'Doctor Ruppel', 'Picardy', 'Fireflame', 'Violet Elizabeth', 'Princess Diana', 'Mrs. Cholmondeley ',' Bagatelle ',' Ball of Flowers'. Experiments on sterilization of plant material have been carried out and aseptic cultures of hard-to-breed clematis varieties have been obtained. The technique of microclonal reproduction has been mastered. On the nutrient media of MurashigeSkug, containing cytokinin 2ip hormones 2ip at a concentration of 3 mg / l, auxin IMC at a concentration of 0.3 mg / l and gibberellin GK3 at a concentration of 1 mg / l, the initiation of proliferation of axillary meristems of aseptic cultures of hard-to-breed clematis varieties was carried out. Various variants of the hormonal composition of the nutrient medium MurashigeSkuga for testing the induction of proliferation of axillary meristems and for mass multiplication of the aseptic clematis culture under in vitro conditions were tested. And the optimal hormonal composition for mass multiplication of clematis is determined. Specific features of morphogenesis of aseptic culture of clematis species depending on the hormonal composition of the nutrient medium are revealed. When cultivating hard-to-breed clematis varieties on variants of MS mediums with different hormonal composition, shoots multiplication was observed on nutrient media with a content of 2ip 1-2 mg / l and BAP 0.5-2 mg / l. At the given concentrations optimal morphogenetic parameters of explants were obtained .
